Somedy suggested that he may have been able to tell Sera is a girl because she doesn't have an Adam's apple (you can see her neck closely on the picture), I think that's possible too.

But wouldn't it make Camel and Jodie just more curious about Sera so that they would investigate Sera? James saw how they reacted to her. So not telling them to not make them both worry sounds strange.

I think James is hidding it not because he wants to be considerate towards Jodie.
Also, James looked a bit creepy when he said that he doesn't know sera XD

I don't think that's the case given that they don't know that he knows who it is. Even if they are suspicious that he may know something, that's even more reason for him to not cave, in hopes that they will forget and let it go (thus protecting them from any further distress). If he tells them, it can only lead them down one path (speaking with Sera, hearing out her doubts, and likely re-opening the "case" on some basis). If he doesn't, it at the very least leaves it likely that they won't stir everything back up again. After all, they don't really have good reason to go after Sera right now--she just appears to be an average teenager that reminds them of "someone". Apart from that, they have no reason to suspect there's any connection to anything greater than that (at this point).

A better counterargument for my money is that James--if he does know--is knowingly letting Sera run wild, inevitably looking into her brother's death. The problem here is that this could potentially put her in danger if the Borg found out. James would know this much, though he may well just suspect she isn't really capable of doing enough to become a target of her own.

Y'know, if they put some hints about Kaito having feelings for Conan, or any other signals of yaoi-ness it could come close to one.

Well, it looks like it really is a tie. 

Heiji managed to pick up a clue that it wasn't suicide. It was actually murder. 

Conan managed to find the clues from the food and revealed the culprit.


Agent Andre Camel managed to meet with his superiors regarding the photo message he sent.

while James did say that about not mentioning Akai, I think there's enough of a difference between something like "I think I saw your dead ex in the bathroom, but he was alive!" and "oh, this girl might be his sister" that it could just as easily be some other reasoning than "don't remind her of Akai" as it could be that reasoning.

I just feel like if the reason for the gender confusion was to lead up to this hint that James knows her, the hint would have more to it than just "James knows Akai's sister" behind it, whatever it may be.
