Any proof the Vermouth uses APTX 4869? I want concrete evidence.

I don't think there's any concrete evidence. Vermouth doesn't look any older after 20 years, but you can't be certain that it is APTX 4869.
I rather not go into theories because there are discussions on that elsewhere.

This is not proof only a question. Remember that it has been proven by fingerprints, that the Mother Sharon is the same person as the Daughter Chris. If this was not done by the drug then how was it done?

vermouth may be at age of 40+ but still there's no proof shown in the anime that she used the 4869...

There is no real evidence that Vermouth used APTX 4869, but there are rumours. As you can see here:

The APTX stands for "Apoptoxin", apop[tosis] means "fall from" in Greek, and toxin is the english word for poison. The drug was created to kill people; the drug should attack the cells and kill them, but instead it makes them shrink. Shinichi and Sherry became 6 year olds again, but Vermouth only got 29 old. There are some rumours, that the drug turns poeple back to the best age of the body. (This is only a rumour, but could be true). So young people become kids, and older people just get younger.

that rumour makes no sense, if the best age for some 50 year old is 29 then isnt the best age for a 16 year old, 16? lol

Vermouth used the secret potion before it was accidentally used for one batch of the apotoxin. The secret potion worked fine on its own, but the other ingredients of the apotoxin made it act up and turn Ai and Conan into kids. 

What secret potion you ask? I'm not at liberty to discuss that information.
