Sera said her elder, second brothers and herself had different surname, their father is dead due to some reason, Sera is their mother's surname, her second brother's surname is Sera until graduated from high school. Then Conan and Sera reacted when Sonoko said "red" (same Japanese words as Akai)

The file is very good, since the apparition of sera, all the files are related to the plot!

Also the background story for the case is really bloody and interesting, usually when the case in itself has those people getting drowned or poisoned it doesn't give off that shock value feeling (because it's purely Mystery most of the time) but when Sera just grabbed the dudes hair and pulled him up it looked pretty neat

This case has some very interesting backstory. But do you think the editor's claim that Sera's backstory will be revealed soon is true, or just a bunch of hot air? I know manga editors tend to know a bit about the current plot arcs before they start publishing, but I'm wondering how much of it is just hype. All we got so far is that she and her brothers all have different names (and at this point we'd already figured that out, even without having to refer to Chek's detailed analyses of the situation). Although, since this is with the Unidentified Akai Brother's classmates, I'm hoping we'll get a little more than just that.

Probably next file we'll get a tiny miniscule hint of plot. Since plot stuff always happen at the beginning and the end in a normal 3 file case.

Right now, Haneda Shuukichi as the middle brother seems plausible, since he is 28. But I just want it to be Amuro. Maybe I like him too much XD

He's also out because her brother isn't supposed to be a detective. On another note, I love it when Sonoko has the habit of being right about stuff she doesn't know about. Just as she said about Haibara that she's like an adult woman in the body of a first grader, she said Akai just Sera mentioned a brother's surname
While Sherlock Holmes is known for his deductive prowess, he acknowledges his brother Mycroft as being superior in that field. However, Mycroft isn't a detective because he lacks the energy for solving cases that Sherlock possesses (as in, he'll give a theory based on the facts, but he won't go out of his way to test or prove it).
I don't know about you guys, but seems to me like there's a similar dynamic between the Akai brothers. On the one hand we have Akai, an FBI agent who's great at deductions. When he makes a theory, he doesn't sit around and wait for others to test or prove it. He was waiting for the BO to show up at Kogoro's with a sniper rifle and took a shot at Gin, and also had people follow Ai after first becoming aware of her resemblance to Shiho. He also faked his death in a manner that reflected Sherlock Holmes, thus making him VERY comparable to Sherlock.
However, as scary as Akai's deduction skills are (and they are VERY scary/impressive), Sera refers to him as the "strong" one and the second brother as the "smart one". This means that this man exceeds Akai's intelligence/deductive skills (which is honestly somewhat hard and scary to imagine). Despite his obviously impressive skills, though, Sera also says that this brother isn't a detective.
Anyone starting to see the resemblance to another fictional detective's brother?
Following this logic (and flipping Mycroft's status as the older brother to the younger brother), then the most likely person for Mycroft is Haneda Shuukichi. Shuukichi is a perfect equivalent for Mycroft: he's great at deductions but isn't a detective, and makes no extra effort to verify his conclusions. Remember the case he appeared in? While I can't remember the exact details off the top of my head, it was at least pretty obvious he figured out the killer's identity but chose not to say anything.
In short, the Akai brothers' relationship seems to strongly reflect that of the Holmes brothers. I don't know whether this was planned by Gosho or just a coincidence (I can't say for sure since the Mycroft-equivalent is the younger brother in DC), but either way, I still think it's pretty interesting to consider.
If it seriously legitly says that Akai is the stronger one, despite we already saw Conan outsmart Amuro and we all saw how powerful Amuros punch was and he even said that his shoulder is broken so he can't "serve as he used to"

It really did surprise me that Akai was the strong one. But, with the parallels with Holmes and Mycroft, I'm reminded of how Conan and Akai have some parallels.
After all, Conan and Akai are comparable in intellectual level to each other. The only one we've seen that matches that is Yuusaku, who is somewhat Conan's equivalent of Mycroft. That narrows the field for the second brother down to a very, very limited range of characters. Shuukichi and Amuro both come to mind but both require some ambiguity on Akai's age.
Akai is lying about being 27 when in disguise, which makes sense given the timeline. To be in the FBI as a field agent, you need to be 23 as the bare minimum. The waiver for age is for qualified people over 37, not under 23. Akai started infiltrating the Black Organization five years before the start of the story. That automatically means he is at least 28 years old. There are certain requirements for being in the FBI as well that could [though not necessarily] make him older yet. You need to be a United States citizen, at least one four year degree from an accredited college/university, and three years work experience before you can join.
The average for applying for and receiving citizenship in the US is at least 5 months, if not longer, too, so that will factor in. On top of that, the FBI's website says it can take anywhere from six months to several years to get into the FBI due to things like the background checks and physical testing. And, given the nature of work with the Black Organization, it's unlikely the FBI would take a fresh special agent without experience as an agent and just throw them to the wolves.
This also gets interesting when one keeps in mind Subaru's quick deduction of who Conan was. As soon as he got more detailed information than 'a high school detective,' he put the pieces together very quickly and confirmed it very quickly through trickery that Conan and Kudou were the same person. If his younger brother is that smart, to the point that Akai is the strong one by comparison, this should get interesting very fast. Shuukichi and Amuro both have suspicions and, from what I remember, neither know much about Shinichi.
Though I will admit that Amuro being the younger one would be an interesting alcohol name pun. Bourbon often replaces Rye in some more recent cocktails, after all. (Speaking of, I should delve into those alcohol puns in more detail in another thread.)

So here it what I have so far starting with the red woman 15 years ago.
Kansuke and Koumei found out by questioning a policeman that he made an error in the case reports. The red woman threw the knife at him in the woods. He continued chasing her, she disappeared, but when he went back for the knife it wasn't there anymore. 3 years later, it was found in very good, non-rusted condition, next to the pond Satoko drowned in with the skeleton of the person who fell in around the time of the red woman attack. Before she drowned Satoko had earlier claimed to have seen the red woman. She was wearing a beige outfit at that time. When her body was found, she was wearing a red coat, and her shoes were nearby. That leads to one reasonable conclusion: Satoko had the knife from three years ago (she took it which explains why it was not rusted out), pretended to see the red woman, dressed up in red, and took off her shoes. She was obviously pretending to play a prank on someone.
It must have backfired on her. In the first chapter of this case, when Ran described the Red woman that she saw, Hakuya thought it was Satoko. That means he was likely the intended victim of Satoko's prank and accidentally killed her, perhaps by knocking her into the bog or throwing something at her so she lost footing like Conan did.
Someone figured out Hakuya killed Satoko, which explains why he became the recent victim. However since Hakuya only just revealed himself, the murder could not have been premeditated. The tomato trick was planned though, like the other pranks that had happened earlier. Since the murderer used the tomato trick as part of the murder, I assumed they also planned the tomato prank. The motive should be either continuing Satoko's pranks, or they were hoping to get someone to confess something.
Based on the fact the murder was not premeditated, it was almost certainly committed by one of the two in the house. That person would need confront the victim to verify the story about Satoko first before committing the murder.
Based on the layout - access to the bathroom where Hakuya works from her cleaning area - Sumiko is the most suspicious. She probably pre-prepared her cleaning chore so she could have extra time to set up the tomato trick. (Maybe she came to the villa in advance or started early.) Everyone has the same chores every year so it's easy to know what they will be asked to do. Sumiko turns on the fan so it is loud and sets it to swivel near a wall. She tapes the bat to the fan so that it bonks periodically and sounds like a vaccuum session in progress.
The trick with the tub has a few possibilities. The water is pretty opaque and there are side areas in the tub you could corral the tomatoes in. If the water is really opaque and not well mixed the culprit could just pour the salt on the bottom. If it is not well mixed, it will create a layer of salt water with a thick layer of fresh on top. The tomatoes won't rise until the salt becomes better mixed throughout the bath.
Sumiko was later attacked by someone who looks like the red woman.
The real red woman is apparently dead: the skeleton was 3 years old, matching the time of the original attacks by The Red Woman Takeno Komayo. The skeleton also genetically matched to a piece of saved umbilical cord that Takeno's mother had.
The red woman who attacked Sumiko may not have been a random murder, or revenge for Hakuya, but instead was one of the housemates trying to remove suspicion from Sumiko by making it appear there is an outsider doing the attacks. Maybe there was something one of the housemates saw that made that person realize that Sumiko was responsible for the tomatoes and thus the murder. Attacking her and giving her a superficial injury would make her seem less suspicious and redirect police attention to outside culprits.
Who is the current red lady?
Why was Hakuya in the bog? Changing clothes? Pretending to be the red lady? Looking to recover evidence like the wig Satoko should have been wearing?
And who's the skeleton? Are we sure it's the dead Red Lady?

I've been suspicious of Sumika ever since she first appear. Unlike the other three, she knew who Sera was without being introduced, which strongly suggests that she was the Red Woman in the forest eavesdropping on their conversation. Is there a possibly that she faked being attacked? (I haven't seen the spoilers for File 784)
